This brings us to the likely crux of the matter. If you cannot play video or audio in a PowerPoint presentation, chances are you are using an unsupported media format.

As for audio, PowerPoint 2010 uses mostly WAV files! For example, if you use an MP3 file in your presentation, you may experience problems.
